PA9/PA10 are available and it is possible to use normal STM32 bootloader through UART, I have used this many times (at least in STM32 version) Unfortunately SWD pins are not available on any other point except SWD connector, but olimex is selling the adapter https://www.olimex.com/Products/ARM/JTAG/...

But it is not necessary to plug/unplug the board very often, assuming that you have spare breadboards.

Try to full erase the chip and reprogram the bootloader only. The enumeration failure is caused in 95% of cases by cable/connector failure or some kind of short circuit in usb lines. Is your MCU OK? Try to program the MCU with serial port and test the behavior of CPU, make some IO operations in USB ...

SysTick is an optional part of Cortex design, but I think that all STM32's even the smallest members of F0x0 they have it.

Specially for the arduino API, I dont find a reason not to use systick. Arduino's millisecond timer is an always-on timer with fixed period.
Systick is not available on all ARM cores but all members of STM32 family have systick, so, it is not problem to use it globally.

There is no problem with L476RG at 80MHz, it is working very well. I have used mbed and Chibios in various small tests with I2C, SPI, UARTS etc and is working... I have also select this MCU (best value/price ratio) for a real project (currently on development) with a custom board.

As I understand, CubeMX passes the parameters on registers as they are (at least for I2C).... For proper operation a rather complex calculation is required, while this is not a big problem, the different parameters for different clocks, is. I will check the mbed and Chibios how they solve this probl...
